Structure

• fenol ring : 2 isopropil
• 10 mg/ml : soy bean oil, glycerol, egg lecithin
• alergi telur biasa putih telur, propofol dari kuning telur

• redistribusi 2-8 menit
• good for outpatient
• elderly : lower dose induction karena Vd smaller pada usia tua
• wanita butuh dosis lebih besar dan bangun lebih cepat

B long term -> lipemia, asidosis metabolite, keratin

• metabolisme ekstrahepatic
• inaktif
• do not affect by moderate cirrhosis

E urine 

• CRF do not affect clearance of the parent drug

EFEK

CV 

• menurunkan aBP karena menurunkan SVR (inhibisi simpatetic vasoconstrictor activity)
• menurunkan kontraksi
• menurunkan preload
• hipotensi lebih sering terjadi daripada thiopental tapi bisa pulih saat laringoskopi intubasi
• sebab hipotensi : dosis besar, injeksi cepat, usia tua
• mengganggu respon baroreflek arteri terhadap hipotensi pada pasien normo dan hipocarbia
• menurunkan preload karena menurunkan HR ; vagal mediated reflex bradicardia
• hati hati pada pasien extreme age, negatif inotropik medication, operasi yang berhubungan dengan refleks okulokardiak

RESP

• resp depresan -> apnea
• inhibit hypoxic ventilatory drive, depress normal response to hypercarbia
• depress upper airway reflex
• histamine release tapi tidak kotraindikasi pada pasien asthma

CEREBRAL

• turunkan CBF, ICP
• propofol dan thipental proteksi fokal iskemia
• antipruritus
• antiemetic
• saat induksi dapat terjadi fenomena eksitasi : muscle twitching, spontan movement, opistotonus, hiccup karena antagonis terhadap subcotical glisin
• antikonvulsan efek
• turunkan IOP
• tdk ada toleransi obat

DRUG INTERACTION (DI)

• dengan NDMR : -. kl dulu potensiasi ndmr krn propofol pake cremophor
• + fentanil dan alfentanil : meningkatkan konsentrasi fentanil dan alfentanil
• dengan midazolam : sinergistik ?
